Doxepin hydrochloride is a dibenzoxepin tricyclic compound. It displays a range of pharmacological actions including maintaining adrenergic innervation. Its mechanism of action is not fully understood, but it appears to block reuptake of monoaminergic neurotransmitters into presynaptic terminals. It also possesses anticholinergic activity and modulates antagonism of histamine h (1) - and h (2)-receptors.
Doxepin hydrochloride is a dibenzoxepin derivative and tricyclic anti- depressant with antipruritic and sedative activities. Doxepin blocks the reuptake of norepinephrine and serotonininto presynaptic terminals thereby prolonging the availability of the monoaminergic neurotran smitters within the synaptic cleft and enhancing their action leading to sedative effects.doxepin also has antagonistic effects on histamine (h1 and h2), 5-ht2, alpha-1 adrenergic, and muscarinic receptors. The antipru- ritic effect of this agent is the result mediated through inhibition of histamine receptors.

doxepin hydrochloride is a white solid powder. It is soluble in water (100 mm), chloroform (1:2), alcohol (1:1), methanol, and dmso with melting range of 188°c to 189°c.
